Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.

In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Tirzepatide is a synthetic peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. The molecule contains 39 amino acids and features a C20 fatty diacid moiety attached via a linker, which promotes albumin binding and extends its circulating half-life. Its sequence incorporates non-natural amino acids and modifications that reduce susceptibility to degradation by dipeptidyl peptidase-4. This dual receptor activity distinguishes it from selective GLP-1 receptor agonists.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
Tirzepatide is a synthetic peptide developed as a dual agonist at the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors. Its structure is built on a GIP-derived backbone with non-natural amino acid substitutions and a fatty diacid side chain that promotes albumin binding and slows clearance. That modification supports once-weekly subcutaneous dosing. Registrational trial programs reported reductions in body weight and glycated hemoglobin alongside the drug's glycemic effects.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Common degradation routes include hydrolysis of labile amide bonds, deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation. Aggregates can form during freeze-thaw cycling, at elevated pH, or when peptide concentration is high. Each route produces characteristic chromatographic or mass shifts that are tracked during stability studies. Whether a given minor impurity alters biological activity is often an open question, and specification limits are typically set on identity and purity rather than on functional data for trace species.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.
Purified material is typically handled as a lyophilized powder kept at or below minus twenty degrees Celsius, shielded from light and moisture. In that state the solid remains stable for extended periods, although repeated freeze-thaw cycling can encourage aggregation. Once dissolved, aqueous solutions are less durable and are generally held cold and used within a brief window. Buffer composition, pH and ionic strength all influence degradation rates, and mildly acidic to neutral conditions are commonly examined. Actual shelf life depends on formulation, concentration and container, so stability limits are established experimentally rather than assumed.
Bagwell disclosed that he was only in good enough condition to play every several days, rather than every day. He was eventually paid the full amount of his contract. The Astros and the insurance company settled the claim in a confidential arrangement the same day he announced his retirement. The Astros declined to pick up Bagwell's $18 million club option for 2007, instead buying it out for $7 million. He filed for free agency in November 2006 but announced his retirement one month later. Owner Drayton McLane and general manager Tim Purpura stated that he would remain in player development department of the Astros organization, as one of the assistants to the general manager.
=== Mechanism of action === Plecanatide works as a laxative by drawing water in to the gastrointestinal tract thereby softening stool and encouraging its natural passage. Similar to its endogenous counterpart, plecanatide activates guanylate cyclase-C on endothelial cells within the gastrointestinal tract. The activation of guanylate cyclase-C catalyses the production of the second messenger guanosine 3’,5’-cyclic monophosphate (cGMP) which leads to the protein kinase A (PKA) and protein kinase G II (PKGII)-mediated phosphorylation of the cystic fibrosis transmembrane conductance regulator (CFTR) protein. CFTR is an anion channel and upon activation it will secrete negatively charged ions, particularly chloride (Cl−) and bicarbonate (HCO3−) in to the GI tract lumen. This disruption to the electrochemical gradient is in part rectified by the passive secretion of positively charged sodium ions in to the lumen and water follows by osmosis. Plecanatide is also known to have an anti-nociceptive effect in animal models, however the exact mechanism of action is not yet fully elucidated. It has been suggested that this may be in part to the anti-inflammatory action of guanylate cyclase-C by its inhibition of pro-inflammatory cytokines, or through the inhibition of associated sensory neurons.
There have been few incidents of doping in football, mainly because of little unannounced testing relative to the high amount of professionals. There is also a lack of further investigation (e.g. Tony Schumacher revelations and the still unknown footballers of Eufemiano Fuentes), preservation of samples, and consequences. In 2014, the biological passport was introduced in the 2014 FIFA World Cup; blood and urine samples from all players before the competition and from two players per team and per match are analysed by the Swiss Laboratory for Doping Analyses.
=== Self-medication theory === The self-medication theory suggests that people with severe mental illnesses misuse substances in order to relieve a specific set of symptoms and counter the negative side-effects of antipsychotic medication. Khantizan proposes that substances are not randomly chosen, but are specifically selected for their effects. For example, using stimulants such as nicotine or amphetamines can be used to combat the sedation that can be caused by higher doses of certain types of antipsychotic medication. Conversely, some people taking medications with a stimulant effect such as the SNRI antidepressants Effexor (venlafaxine) or Wellbutrin (bupropion) may seek out benzodiazepines or opioid narcotics to counter the anxiety and insomnia that such medications sometimes evoke. Some studies show that nicotine administration can be effective for reducing motor side-effects of antipsychotics, with both bradykinesia (stiff muscles) and dyskinesia (involuntary movement) being prevented.
Fe(II) + H2O2 → Fe(III)OH + HO· The Fenton reaction explains the toxicity of hydrogen peroxides because the hydroxyl radicals rapidly and irreversibly oxidize all organic compounds, including proteins, membrane lipids, and DNA. Hydrogen peroxide is a significant source of oxidative DNA damage in living cells. DNA damage includes formation of 8-Oxo-2'-deoxyguanosine among many other altered bases, as well as strand breaks, inter-strand crosslinks, and deoxyribose damage. By interacting with Cl−, hydrogen peroxide also leads to chlorinated DNA bases. Hydroxyl radicals readily damage vital cellular components, especially those of the mitochondria. The compound is a major factor implicated in the free-radical theory of aging, based on its ready conversion into a hydroxyl radical.

=== Nutritional displacement === The "empty calories" argument states that a diet high in added (or 'free') sugars will reduce consumption of foods that contain essential nutrients. This nutrient displacement occurs if sugar makes up more than 25% of daily energy intake, a proportion associated with poor diet quality and risk of obesity. Displacement may occur at lower levels of consumption.
=== August === 1 August – A BBC News investigation discovers that people earning more than £30,000 are being told they are not eligible to rent affordable housing in London because they do not earn enough money. 3 August – Rashid Gedel, 22, and Shiroh Ambersley, 22, are sentenced to life imprisonment with a minimum term of 27 years for the February 2021 murder of Sven Badzak in North West London, who was stabbed in a case of mistaken identity. Gedel would later allegedly murder convicted pedophile and former Lostprophets frontman Ian Watkins in prison in October 2025. 4 August – The UK government announces plans to tackle the NHS backlog with the opening of 13 new regional diagnostic centres, eight of which will be privately run. The government also wants to make it easier for the NHS to access services from the private sector. Following a trial at Derby Crown Court, Craig Crouch is sentenced to life imprisonment with a minimum term of 28 years for the murder of his 10-month-old stepson, Jacob Crouch, in December 2020. Jacob's mother, Gemma Barton, is sentenced to 10 years imprisonment for causing or allowing the death of a child. The scrappage scheme for non-compliant vehicles is extended to the whole of London ahead of the expansion of the Ultra Low Emission Zone. Following a trial at Leicester Crown Court, TikTok social influencer Mahek Bukhari and her mother Ansreen Bukhari, are found guilty of murdering two men whose car was rammed off the road following a dispute over an affair Mrs Bukhari had been having with one of the men.
== Tributes == "Referring to the death of Professor Brailsford Robertson yesterday, the Prime Minister (Mr. Scullin) said:— His untimely end has robbed Australia of a brilliant scientist, whose contribution to the Commonwealth's progress would always be remembered. On behalf of the Federal Ministry Mr. Scullin extended his sincere sympathy to Professor Robertson's family, and expressed its appreciation of the great national services which he had rendered." The Argus, 20 January 1930. "Professor Brailsford Robertson was one of Australia's most brilliant scientists. The value of the work he was accomplishing cannot be estimated." Hon. R.L. Butler, Premier of South Australia, 20 January 1930. "The main purpose of [Robertson's] studies was to solve the mystery of physical life in it successive stages of commencement, growth, decline, and cessation, for the benefit of his fellow-men. ... The amount of original work he accomplished in his short life was prodigious. I would be rash to assert that the loss to science, or as I should prefer to say, the loss to humanity, caused by his death is irreparable, but it will probably be a long time before anyone else is able to gather the threads together and produce the fabric he was attempting to weave on his loom." Sir George Murray, Chancellor of the University of Adelaide, 18 March 1932.
McGuinn's concept was jettisoned in favor of a country project, which included Parsons' songs "One Hundred Years from Now" and "Hickory Wind", and songs by Bob Dylan, Woody Guthrie and Merle Haggard. Recording sessions for Sweetheart of the Rodeo commenced at Columbia Records' recording studios in the Music Row area of Nashville on March 9, 1968. Mid-way through, the sessions moved to Columbia Studios, Hollywood and were completed on May 27, 1968. However, Parsons was still under contract to LHI Records and Hazlewood threatened legal action over Parsons' appearance on the album. As a result, McGuinn replaced three of Parsons' lead vocals with his own; in 1973, in an interview with Cameron Crowe, Parsons said that McGuinn "erased it and did the vocals himself and fucked it up." Parsons remained as lead vocalist on the songs "You're Still on My Mind", "Life in Prison", and "Hickory Wind". While in England with the Byrds in the summer of 1968, Parsons left the band over a planned concert tour of South Africa and his opposition to apartheid. During this period Parsons became acquainted with Mick Jagger and Keith Richards of The Rolling Stones. After leaving the band, Parsons stayed at Richards' house and the pair developed a close friendship, with Parsons reintroducing the guitarist to country music.
==== Neurological ==== Peripheral neuropathy and mononeuritis multiplex may occur. The most common problem is carpal tunnel syndrome caused by compression of the median nerve by swelling around the wrist. Rheumatoid disease of the spine can lead to myelopathy. Atlanto-axial subluxation can occur, owing to erosion of the odontoid process or transverse ligaments in the cervical spine connection to the skull. Such an erosion (>3mm) can give rise to vertebrae slipping over one another and compressing the spinal cord. Clumsiness is initially experienced, but without due care, this can progress to quadriplegia or even death. Vertigo may be associated with rheumatoid arthritis via the following associations that can cause vertigo:
