Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
As a peptide, tirzepatide is handled as a lyophilised solid in research settings and as a preserved solution in finished products. Aqueous solubility is pH dependent and reaches a minimum near the isoelectric point, which lies close to pH 5.4. Stock solutions are typically prepared in neutral or slightly basic buffer to limit precipitation. The solid is hygroscopic and should be equilibrated to room temperature before opening so that condensation does not form on the powder surface.

Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.
After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
Tirzepatide is a synthetic peptide of 39 amino acids engineered from the native glucose-dependent insulinotropic polypeptide sequence. Its structure incorporates several non-natural residues and a C-terminal segment derived from glucagon-like peptide-1, together with a C20 fatty diacid moiety attached through a linker. The lipophilic side chain promotes binding to serum albumin, which slows renal clearance after administration. The compound is classified as a dual incretin receptor agonist and is supplied as a lyophilized powder for reconstitution or as a preformulated solution, depending on the presentation.
Purified material is typically handled as a lyophilized powder kept at or below minus twenty degrees Celsius, shielded from light and moisture. In that state the solid remains stable for extended periods, although repeated freeze-thaw cycling can encourage aggregation. Once dissolved, aqueous solutions are less durable and are generally held cold and used within a brief window. Buffer composition, pH and ionic strength all influence degradation rates, and mildly acidic to neutral conditions are commonly examined. Actual shelf life depends on formulation, concentration and container, so stability limits are established experimentally rather than assumed.
Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.
The compound first appeared in the scientific literature as an investigational agent for type 2 diabetes. Clinical development proceeded through phase 1, phase 2, and phase 3 programs that measured glycemic control as a primary endpoint while recording body weight as a secondary outcome. Regulatory approval in the United States followed in 2022 for glycemic control, and a separate indication for chronic weight management was added later. Subsequent trials have examined cardiovascular outcomes in adults with elevated cardiovascular risk. Debates continue over how much of the observed effect derives from each receptor arm.
CGRP is released from both the trigeminal ganglion (TG) and the trigeminal nucleus caudalis (TNC) in response to trigeminal nerve activation. CGRP activates receptors on meningeal blood vessels, causing dilation and changes in blood flow. CGRP also activates specialized nerve endings on the dura mater (nociceptors) that transmit pain signals from the dura to the central nervous system. Increased neuronal activity in the trigeminal pain pathway reaches higher cortical pain regions via the brainstem, midbrain and thalamus. Stimulation of the trigeminal nerve may result in release of neuropeptides such as CGRP, vasodilation of cerebral and dural blood vessels, neurogenic inflammation, and the transmission of pain signals via nerves in the meninges. Cerebrospinal fluid may also play a role in migraine by transferring signals released from the brain to overlying pain-sensitive meningeal tissues, including dura mater.
== Preparation == Since its first description in 1953, ethylene carbonate has been commonly used as starting material for vinylene carbonate. In the first stage, monochlorethylene carbonate is produced in a UV-initiated photochlorination reaction with chlorine or sulfuryl chloride at 60-70 °C in bulk. In the second stage, monochlorethylenecarbonate undergoes dehydrochlorination with a base such as triethylamine.
== Actions and psychopharmacology == CRH is produced in response to stress, predominantly by parvocellular neurosecretory cells within the paraventricular nucleus of the hypothalamus and is released at the median eminence from neurosecretory terminals of these neurons into the primary capillary plexus of the hypothalamo-hypophyseal portal system. The portal system carries the CRH to the anterior lobe of the pituitary, where it stimulates corticotropes to secrete adrenocorticotropic hormone (ACTH) and other biologically-active substances (β-endorphin). ACTH stimulates the synthesis of cortisol, glucocorticoids, mineralocorticoids and DHEA. In the short term, CRH can suppress appetite, increase subjective feelings of anxiety, and perform other functions like boosting attention. During chronic stress conditions such as post-traumatic stress disorder (PTSD), blood serum levels of CRH are decreased in combat veterans with PTSD compared to healthy individuals. It is believed that chronic stress enhances the negative feedback inhibition of the HPA axis, resulting in lower CRH levels and HPA function. Abnormally high levels of CRH have been found in people with major depression, and in the cerebrospinal fluid of people who have committed suicide. Corticotropin-releasing hormone has been shown to interact with its receptors, corticotropin-releasing hormone receptor 1 (CRFR1) and corticotropin-releasing hormone receptor 2 (CRFR2), in order to induce its effects.
Crabs (Carcinus maenas) that were fed a diet of mussels contaminated with environmentally realistic concentrations of branched alkylbenzenes, suffered behavioural disruption but only a small concentration of the compounds were retained in the midgut of the crabs. Within marsh sediments still contaminated with high concentrations of UCM hydrocarbons from the Florida barge oil spill in 1969 (see above,) the behaviour and feeding of fiddler crabs (Uca pugnax) was reported to be affected.
Hardtack (or hard tack) is a type of dense cracker made from flour and small amounts of water, with salt being sometimes added. Hardtack is very inexpensive and long-lasting if kept dry, allowing it to be used for sustenance in the absence of perishable foods. It is commonly used during long sea voyages, land migrations, and military campaigns. Along with salt pork and corned beef, hardtack was a standard ration for many militaries and navies from the 17th to the early 20th centuries.

=== Japan === Manfred Kiese at the Pharmacological Institute in Berlin published a survey of literature on antibiotics in the 7 August 1943 issue of Klinische Wochenschrift that included the Oxford team's publications. A copy was acquired by the Japanese embassy in Berlin and taken to Japan on the Japanese submarine I-8, which docked at Kure, Hiroshima, on 21 December 1943. The article was translated into Japanese, and production of penicillin was underway by 1 February 1944. By mid-May, a research team under Hamao Umezawa had tested 750 different strains of mould and found that 75 exhibited antibiotic activity. Experiments were conducted on mice to determine efficacy and toxicity. The Morinaga Milk company had a small penicillin production plant in operation in Mishima, Shizuoka, by the end of the year, and the Banyu Pharmaceutical Company opened a small plant in Okazaki, Aichi, in January 1945. The penicillin was called "Hekiso" after its blue colour. By 1948 Japan had become the third country, after the US and UK, to become self-sufficient in penicillin, and exports to China and Korea began the following year.
Buc-ee's has a beaver mascot named Buc-ee that it uses for various promotional and merchandising purposes. He is an anthropomorphic beaver wearing a baseball cap and t-shirt. The character's design in Buc-ee's's logo was made using a drawing guide from Preston Blair's book Advanced Animation. Statues of Buc-ee can be found in many locations of the gas station, and one is even displayed at a museum in Austin, Texas. Buc-ee also has a mascot costume, which is used for promotional materials and to advertise store openings. Buc-ee's is very litigious when it comes to their mascot, as they have filed several lawsuits against companies they deem to be infringing on his design.
Rpn11 is an intrinsic, stoichiometric subunit of the 19S regulatory particle and is essential for the function of 26S proteasome. Rpn11 is a zinc-dependent, metalloprotease of the JAB1/MPN/Mov34 metalloenzyme (JAMM) family of DUBs, that was identified to be the essential DUB responsible for the en block removal of the ubiquitin chain from the protein substrate. Rpn11 forms an obligate dimer with Rpn8 forming an active DUB able to cleave all ubiquitin linkages. The active site of Rpn11 is formed through metal coordination of the catalytic zinc and this site is covered by an Insert-1 loop that covers this active site. The structure is very similar to that of a related JAMM DUB, AMSH, that is responsible for K63 ubiquitin cleavage, however it lacks the residues that are key for AMSH's linkage specificity. The structure of Rpn11 bound to ubiquitin revealed that the C-terminus of Ubiquitin pushes the insert-1 loop into an beta-sheet providing access to the catalytic zinc. This structure combined with detailed biochemistry revealed that the DUB activity of Rpn11 was accelerated at least 10-fold by the translocation of the protein substrate, suggesting that the translocation delivered the Ub substrate to the active site of Rpn11. This model of translocation-dependent deubiquitination was later confirmed by cryoEM of both the yeast and human proteasome bound to a substrate, both of which recapitulated the crystal structure of Ubiquitin bound to Rpn11.

On 12 November 1935 at the Hospital de Santa Marta in Lisbon, Moniz initiated the first of a series of operations on the brains of people with mental illnesses. The initial patients selected for the operation were provided by the medical director of Lisbon's Miguel Bombarda Mental Hospital, José de Matos Sobral Cid. As Moniz lacked training in neurosurgery and his hands were impaired by gout, the procedure was performed under general anaesthetic by Pedro Almeida Lima, who had previously assisted Moniz with his research on cerebral angiography. The intention was to remove some of the long fibres that connected the frontal lobes to other major brain centres. To this end, it was decided that Lima would trephine into the side of the skull and then inject ethanol into the "subcortical white matter of the prefrontal area" so as to destroy the connecting fibres, or association tracts, and create what Moniz termed a "frontal barrier". After the first operation was complete, Moniz considered it a success and, observing that the patient's depression had been relieved, he declared her "cured" although she was never, in fact, discharged from the mental hospital. Moniz and Lima persisted with this method of injecting alcohol into the frontal lobes for the next seven patients, but, after having to inject some patients on numerous occasions to elicit what they considered a favourable result, they modified the means by which they would section the frontal lobes.
An enlarged prostate is called prostatomegaly, with benign prostatic hyperplasia (BPH) being the most common cause. BPH refers to an enlargement of the prostate due to an increase in the number of cells that make up the prostate (hyperplasia) from a cause that is not a malignancy. It is very common in older men. It is often diagnosed when the prostate has enlarged to the point where urination becomes difficult. Symptoms include needing to urinate often (urinary frequency) or taking a while to get started (urinary hesitancy). If the prostate grows too large, it may constrict the urethra and impede the flow of urine, making urination painful and difficult, or in extreme cases completely impossible, causing urinary retention. Over time, chronic retention may cause the bladder to become larger and cause a backflow of urine into the kidneys (hydronephrosis). BPH can be treated with medication, a minimally invasive procedure or, in extreme cases, surgery that removes the prostate. In general, treatment often begins with an alpha-1 adrenergic receptor antagonist medication such as tamsulosin, which reduces the tone of the smooth muscle found in the urethra that passes through the prostate, making it easier for urine to pass through. For people with persistent symptoms, procedures may be considered. The surgery most often used in such cases is transurethral resection of the prostate, in which an instrument is inserted through the urethra to remove prostate tissue that is pressing against the upper part of the urethra and restricting the flow of urine.
=== Detection in body fluids === Stanozolol is subject to extensive hepatic biotransformation by a variety of enzymatic pathways. The primary metabolites are unique to stanozolol and are detectable in the urine for up to 10 days after a single 5–10 mg oral dose. Methods for detection in urine specimens usually involve gas chromatography-mass spectrometry or liquid chromatography-mass spectrometry.
7 June Dan Bullock was the youngest U.S. serviceman to be killed in the war at age 15. Having lied about his age to join the Marine Corps he was killed in a sapper attack at An Hoa Combat Base. Following a rocket and artillery barrage, at 04:30 two battalions of the PAVN 88th Regiment attacked Firebase Crook, but were forced back by gunship fire. PAVN losses were 323 killed without loss among U.S. forces.
Department of Defense in 2012, when the devices were not yet commercially available and therefore did not require FDA approval. FDA inspection reports from 2014 and 2015 stated that its containers for blood collection were "not validated under actual or simulated use conditions" and "were not reviewed and not approved by designated individual(s) prior to issuance". In 2015, an FDA inspection resulted in multiple observed violations of FDA Title 21 Regulations. It was eventually revealed that the FDA had classified Theranos's device, called a nanotainer, as a Class II medical device, meaning that Theranos would need to use special labels, meet certain performance standards and perform post-market surveillance of the device. Theranos asserted that the nanotainer was a Class I medical device and therefore not subject to any regulatory requirements. After the 2015 inspection, Theranos announced that it would voluntarily suspend its tests apart from the FDA-approved herpes simplex virus (HSV-1) test. The Arizona Department of Health Services reported issues with the company's Scottsdale lab meeting regulations in September 2015. The reports were revealed in the Arizona Republic in November 2015. In January 2016, the Centers for Medicare and Medicaid Services (CMS) sent a letter to Theranos based on a 2015 inspection of its Newark, California lab, reporting that the facility caused "immediate jeopardy to patient health and safety" due to a test to determine the correct dose of the blood-thinning drug warfarin.
